Why 165 points is the target
Each of the six branches has a main skill and two subskills. PowerPyx gives the total branch costs as 15, 20, 25, 30, 35 and 40 points, which add to 165.
The order grows more expensive as you buy branches, but the final combined cost is the same. With 200 points available, you can miss 35 and still unlock every skill.

The missable save behavior
A pickup saves at the next checkpoint. If Sophia dies before reaching it, collect the point again. Replaying an older chapter restores the point state attached to that chapter, so farming an old area will not combine with a later total.
If you miss a large cluster, restarting that chapter immediately is safer than finishing the campaign and hoping to repair the point total afterward.

Does buying only the large node finish a branch?
No. The two subskills also count toward completing each mastery branch.
